چکیده

The proton magnetic resonance spectrum of DL-a!-lecithin at 100 MHz in CHCJ solvent is reported. An analysis of the spectrum is made. The chemical shifts of the N-(CH& protons and the resonance peak associated with the condensed water molecule showed concentration dependence with the latter showing the more pronounced response. These chemical shift changes are interpreted in terms of micelle formation through intermolecular hydrogen bond formation between lecithin monomers. The chemical shift data are used to calculate the critical micelle concentration of lecithin as well as the number of monomer lecithin units required to form a micelle. The temperature-dependent studies showed a small negative enthalpy change associated with the micelle. The micellar behavior in CHCl, is compared with those in benzene and carbon tetrachloride.

Techniques and applications of NMR to membrane proteins.

The fact that membrane proteins are notoriously difficult to analyse using standard protocols for atomic-resolution structure determination methods have motivated adaptation of these techniques to membrane protein studies as well as development of new technologies. With this motivation, liquid-state nuclear magnetic resonance (NMR) has recently been used with success for studies of peptides and...

Solution structure of the integral human membrane protein VDAC-1 in detergent micelles.

The voltage-dependent anion channel (VDAC) mediates trafficking of small molecules and ions across the eukaryotic outer mitochondrial membrane. VDAC also interacts with antiapoptotic proteins from the Bcl-2 family, and this interaction inhibits release of apoptogenic proteins from the mitochondrion. We present the nuclear magnetic resonance (NMR) solution structure of recombinant human VDAC-1 r...

Optimization of bicelle lipid composition and temperature for EPR spectroscopy of aligned membranes.

We have optimized the magnetic alignment of phospholipid bilayered micelles (bicelles) for EPR spectroscopy, by varying lipid composition and temperature. Bicelles have been extensively used in NMR spectroscopy for several decades, in order to obtain aligned samples in a near-native membrane environment and take advantage of the intrinsic sensitivity of magnetic resonance to molecular orientati...

Transverse relaxation-optimized NMR spectroscopy with the outer membrane protein OmpX in dihexanoyl phosphatidylcholine micelles.

The (2)H,(13)C,(15)N-labeled, 148-residue integral membrane protein OmpX from Escherichia coli was reconstituted with dihexanoyl phosphatidylcholine (DHPC) in mixed micelles of molecular mass of about 60 kDa. Transverse relaxation-optimized spectroscopy (TROSY)-type triple resonance NMR experiments and TROSY-type nuclear Overhauser enhancement spectra were recorded in 2 mM aqueous solutions of ...
